چکیده

Background: HSV-2, one of the most common HIV-1 copathogens, establishes a synergistic interaction with HIV-1. Several clinical trials have shown that the anti herpetic drug acyclovir decreases HIV-1 viral load and delays HIV-1 disease progression. Similarly, CAPRISA004 reported that a vaginal gel containing 1% of the anti-HIV-1 drug tenofovir unexpectedly decreases the risk of HSV-2 acquisition by 51%. Using a system of cervicovaginal tissue culture that reflects many of the in vivo tissue features, we evaluated the dual anti-HIV-1/HSV-2 activity of acyclovir and tenofovir.

Interleukin-7 Facilitates HIV-1 Transmission to Cervico-Vaginal Tissue ex vivo

The majority of HIV-1 infections in women occur through vaginal intercourse, in which virus-containing semen is deposited on the cervico-vaginal mucosa. Semen is more than a mere carrier of HIV-1, since it contains many biological factors, in particular cytokines, that may affect HIV-1 transmission. The concentration of interleukin (IL)-7, one of the most prominent cytokines in semen of healthy...

Topical tenofovir, a microbicide effective against HIV, inhibits herpes simplex virus-2 replication.

The HIV reverse-transcriptase inhibitor, tenofovir, was recently formulated into a vaginal gel for use as a microbicide. In human trials, a 1% tenofovir gel inhibited HIV sexual transmission by 39% and, surprisingly, herpes simplex virus-2 (HSV-2) transmission by 51%. We demonstrate that the concentration achieved intravaginally with a 1% tenofovir topical gel has direct antiherpetic activity. ...

Chitosan and Kappa-Carrageenan Vaginal Acyclovir Formulations for Prevention of Genital Herpes. In Vitro and Ex Vivo Evaluation

Vaginal formulations for the prevention of sexually transmitted infections are currently gaining importance in drug development. Polysaccharides, such as chitosan and carrageenan, which have good binding capacity with mucosal tissues, are now included in vaginal delivery systems. Marine polymer-based vaginal mucoadhesive solid formulations have been developed for the controlled release of acycl...

Uterus Dysplasia Associated with Cervico-Vaginal Agenesis

Müllerian ducts can form upper parts of normal female reproductive system and any failure in ductal fusion may lead to Müllerian Duct Anomalies (MDA). We present a case of MDA, a uterus dysplasia with no evidence of cervical or upper vaginal tissue. This case illustrates the role of Magnetic Resonance Imaging (MRI) on MDA diagnosis and urges the need for a unified reliable and practical classif...

Vaginal Lactobacillus Inhibits HIV-1 Replication in Human Tissues Ex Vivo

Lactobacillus species, which dominate vaginal microbiota of healthy reproductive-age women, lower the risks of sexually transmitted infections, including the risk of human immunodeficiency virus (HIV) acquisition. The exact mechanisms of this protection remain to be understood. Here, we investigated these mechanisms in the context of human cervico-vaginal and lymphoid tissues ex vivo. We found ...
